Is it possible to assign a hot key or set a macro up to activate the sample plot (in the hamburger menu "Audio Data")? I turn it on and off quite often and would prefer to click a single button rather tham drill down 3 clicks.

Setup and save timeline views with and without sample plot. Assign those timelines to different workspaces and map the workspaces to your KB
Try menu to button assignment and drag sample plot to a key. (have not tried this as workspaces work for me)
I've got mine mapped to F7. Look up "Menu to Button" mapping in the Help/Manual.

Yup, what you need is "Menu To Button Reassignment"

That worked great, I now have the "SP" button on the timeline, thank you. I don't think I'd have figured it out without asking (not quickly anyway ;).
